Ewing's sarcoma is a childhood/young adult cancer. Unfortunately, many Ewing's sarcoma cancer patients are first diagnosed when the cancer cells are already spreading throughout the body, and these patients have a very poor prognosis. Therefore, identifying novel means to improve the effectiveness of the standard treatment of this cancer, a cocktail of drugs that includes etoposide, would be of immediate benefit. This proposal will provide the basis for novel strategies in the treatment of human cancer.

The proposed Cancer Biology Training Program is a resubmission of an application for an institutional training grant. We request funds to support 2 predoctoral and 2 postdoctoral trainees. Despite all the advances made in cancer research, it has become the number one killer of people under 85 in the U.S. The goal of the Cancer Biology Training Program is to educate the next generation of cancer researchers to meet the growing demands for scientists trained in the multiple facets of cancer biology.

The long latency in clinically significant prostate cancer development provides numerous intervention opportunities. The existence of oxidative stress markers in prostate cancer patients suggests that oxidative stress signaling may be a reasonable target to prevent clinical prostate cancer. Although the failure of the SELECT trial argues against this approach; we propose that a cocktail of antioxidants with the capacity to neutralize several classes of reactive species would be an excellent prophylactic remedy.

Prostate cancer is the most common non-dermatologic neoplasm in men, affecting about one man in six in his lifetime. The primary public health approach for control of this disease is currently early diagnosis and treatment, relying primarily on the Prostate Specific Antigen (PSA) blood test for detection. Unfortunately, for most men with a PSA above 4.0 ng/mL (the most commonly-applied upper limit of normal), no prostate cancer is found at biopsy while many cancers are found below this 'normal' level.

This grant proposes the development of a novel methodology, based on computational approaches and experiments performed under physiological solution conditions, to complement high-resolution methods for the structure determination of biological macromolecules and their assemblies, and the application of this methodology to two relevant cases, Hsp70 and TGFø. Starting with high-resolution structures, the developed software will generate alternative conformations using molecular dynamics. The conformations will be grouped into equivalence classes (ECs).

Proinflammatory cytokines act in the brain to regulate important biobehavioral processes that are critical for physical and mental health. Patients receiving cytokine immunotherapy for treatment of viral infections or malignancies and those suffering from chronic inflammatory diseases are at an increased risk of developing depressive disorders. The neurobiological mechanisms and immune substrates underlying these immune-brain interactions remain unclear. Acute and chronic activation of the peripheral innate immune system in laboratory rodents precipitates depressive-like biobehaviors.
